기존의 Chemiluminescent Western에서 secondary antibody를 IR Dye가 Label된 것으로 변경하고 Li-Cor사에서 제공하는 Odyssey CLx 혹은 Odyssey Fc를 사용하게 되면, Chemiluminescent Western결과에 비해 실험시간과 비용을 절약할 수 있으며, 미량의 시료로 인한 약한 signal도 보다 더 잘 확인할 수 있습니다. No ECL → ECL 비용 절감 No 필름, No 암실 No 시간에 따라 Signal 변화 →항상 일정한 Signal, 필요시 몇 달 후에 다시 Imaging 가능 No Stripping & Reprobing →한 Membrane에서 two target 동시에 검출, 분석. 편리한 normalization No Protocol 변경의 어려움 →이차 항체만 IRDye labeled secondary antibody 이용하며 기존 protocol 그대로 이용
◎Odyssey series를 이용하면 실험시간 및 스텝이 절약됨 Membranetransfer한 이후 Primary antibody를 붙이는 단계까지는 기존 Chemiluminescent western 방법과 동일하며, 이후 IRDye가 labled된 secondary antibody를 붙인 이후 Odyssey Imaging System으로 image를 얻어면 됩니다. IRDye가 labed된 secondary antibody를 붙인 이후 실험 과정은 chemiluminescent 방법 보다 실험 step을 줄일 수 있어, 시간과 비용을 절약할 수 있습니다.
